DETEKSI SENYAWA LOVASTATIN PADA KULTUR MISELIUM DAN TUBUH BUAH JAMUR TIRAM (Pleurotus ostreatus)

ABSTRACT


Fungi from the Pleurotus genus has been reported to produce lovastatin, a potential secondary metabolite as an anti-hypercholesterolemic agent (Chang dan Miles,1973). This research was aimed to study whether lovastatin was produced in the mycelial of P. ostreatus and to measure its quantity. The essay was conducted to detect lovastatin towards two types of samples, which are (A) filtrate from submerged culture of P.ostreatus mycelial (B) filtrate of P.ostreatus mycelial fermentation by L. fermentum. Submerged culture of the mycelia was conducted in potato dextrose broth and fermentation was in sugar solution. Thin layer chromatography (TLC) method was used for lovastatin detection. The eluent used in TLC method was a mixture of dichloromethane and ethyl acetate (70:30,v/v) and acetonitrile as the solvent (Samiee,2003). Lovastatin standard produced a purlple band with the Rf value of 0.87. Qualitative analysis showed that both sample A and B had the same color band and Rf value with the standard. Quantitative analysis resulted that in sample A there was 9.3 mg/L of lovastatin while in sample B there was 24.3 mg/L. From this research it can be concluded that lovastatin was produced by the mycelial vegetative culture of P.ostreatus.
